Product Overview
This rabbit polyclonal antibody detects protein-arginine deiminase type-3 (gene PADI3) in human, mouse and rat samples and is validated for Western blot, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 74.7 kDa, and the supplier reports an observed band at about 75 kDa.
PADI3 (PAD3) is a Ca2+-dependent peptidylarginine deiminase that converts protein arginine residues to citrulline. It is expressed mainly in hair follicles and epidermis, where citrullination of structural proteins such as trichohyalin supports hair shaft formation.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human, Mouse, Rat) |
|---|---|
| Immunocytochemistry / Immunofluorescence | 5 µg/mL (Human) |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human RT4 cells, human A549 cells, rat C6 cells, mouse kidney tissue, mouse NIH/3T3 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (ICC/IF): HeLa cells.
Samples with a confirmed signal (flow cytometry): SH-SY5Y cells.
Immunogen
Recombinant human PADI3 fragment (Glu24–Arg557), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 85.8% and 86.5%, respectively.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places protein-arginine deiminase type-3 in the cytoplasm. Tissue expression noted by UniProt (human): Hair follicles, and epidermis at very low levels. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
