Product Overview

This rabbit polyclonal antibody detects palladin (gene PALLD) in human samples and is validated for Western blot and ELISA. The predicted molecular weight is 150.6 kDa, and the supplier reports an observed band at about 95 kDa.

Palladin (PALLD) is an actin-associated scaffold protein found in stress fibers, focal adhesions and lamellipodia. It binds several actin-binding partners, including α-actinin, and helps organize actin bundles that control cell shape, adhesion and migration.

Host / Clonality / Isotype Rabbit / Polyclonal / IgG
Reactivity Human
Form Lyophilized
Formulation Per vial: 4 mg trehalose; 0.9 mg NaCl; 0.2 mg Na2HPO4
Calculated MW 150.6 kDa

Storage As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.

Validated Applications

Western blot 0.25–0.5 µg/mL (Human)
ELISA 0.1–0.5 µg/mL

Samples with a confirmed band (WB): human HeLa cells, human A549 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Immunogen

Recombinant human Palladin/PALLD fragment (His89–Leu1383), expressed in E. coli. Sequence identity with the mouse ortholog: 85.5%.

Reactivity Notes

The supplier lists reactivity with human. UniProt places palladin in the cytoplasm, cell junction and cell projection. Tissue expression noted by UniProt (human): Detected in both muscle and non-muscle tissues. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

For research use only; not for diagnostic or therapeutic procedures. Handle with standard laboratory precautions.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.25–0.5 µg/mL (Human); ELISA 0.1–0.5 µg/mL. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 150.6 kDa; observed band at about 95 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Recombinant human Palladin/PALLD fragment (His89–Leu1383), expressed in E. coli. Sequence identity with the mouse ortholog: 85.5%.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
