Product Overview
This rabbit polyclonal antibody detects protein mono-ADP-ribosyltransferase PARP10 in human samples and is validated for Western blot, flow cytometry and ELISA. The predicted molecular weight is 110.0 kDa, and the supplier reports an observed band at about 140 kDa.
PARP10 is a mono-ADP-ribosyltransferase that attaches single ADP-ribose groups to target proteins. It interacts with PCNA and supports DNA damage tolerance during replication, and it has been linked to regulation of GSK3B and NF-κB signaling.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human) |
|---|---|
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human Jurkat cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with a confirmed signal (flow cytometry): MCF-7 cells.
Immunogen
Recombinant human PARP10 fragment (Glu14–Glu251), expressed in E. coli. Sequence identity with the mouse ortholog: 76.5%.
Reactivity Notes
The supplier lists reactivity with human. UniProt places protein mono-ADP-ribosyltransferase PARP10 in the nucleus and cytoplasm. Tissue expression noted by UniProt (human): Highly expressed in spleen and thymus. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
