Product Overview
This rabbit polyclonal antibody targets pterin-4-alpha-carbinolamine dehydratase (gene PCBD1) in human and mouse samples and is validated for Western blot, ICC/IF and ELISA. The predicted molecular weight is 12.0 kDa, and the supplier reports an observed band at about 12 kDa.
PCBD1 is a bifunctional protein. In the cytoplasm it acts as pterin-4α-carbinolamine dehydratase, helping regenerate the tetrahydrobiopterin cofactor, while in the nucleus it binds HNF1A and HNF1B and boosts their transcriptional activity (hence its alias DCoH).

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human, Mouse) |
|---|---|
| Immunocytochemistry / Immunofluorescence | 5 µg/mL (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human Caco-2 cells, human HepG2 cells, mouse pancreas tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (ICC/IF): HeLa cells.
Immunogen
Recombinant human PCBD1 fragment (Met1–Thr105), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 99% and 100%, respectively.
Reactivity Notes
The supplier lists reactivity with human and mouse. UniProt places pterin-4-alpha-carbinolamine dehydratase in the cytoplasm and nucleus. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
