Overview

This antibody is intended for detection of PCDH15 (Breast cancer metastasis-suppressor 1) in biological samples using common immunoassay formats. It is typically selected based on target identity, species reactivity, clonality/clone information, and detection modality.

Vendor notes: Boster Bio Anti-PCDH15 Antibody Picoband® catalog # A03591-1. Tested in Flow Cytometry, IHC, IHC-F, ICC, WB applications. This antibody reacts with Human, Mouse, Rat. The brand Picoband indicates this is a premium antibody that guarantees superior quality, high affinity, and strong signals with minimal background in Western blot applications. Only our best-performing antibodies are designated as Picoband, ensuring unmatched performance.

Key elements and design rationale

  • Antibody format: Rabbit Polyclonal Rabbit IgG
  • Immunogen / epitope context: A synthetic peptide corresponding to a sequence at the C-terminus of human PCDH15, identical to the related mouse and rat sequences.
  • Molecular weight context: reported MW: 190 kDa; calculated MW: 28461 MW
  • Reactivity: Human,Mouse,Rat
  • Applications: Flow Cytometry, IHC, IHC-F, ICC, WB

As a polyclonal antibody, the reagent recognizes multiple epitopes on the target, which can improve detection robustness but may increase sensitivity to sample-dependent epitope changes.

Biological background

Breast cancer metastasis-suppressor 1; protocadherin related 15. Protocadherin-15 is a protein that in humans is encoded by the PCDH15 gene. This gene is mapped to 10q21.1. This gene is a member of the cadherin superfamily. Family members encode integral membrane proteins that mediate calcium-dependent cell-cell adhesion. It plays an essential role in maintenance of normal retinal and cochlear function. Mutations in this gene result in hearing loss and Usher Syndrome Type IF (USH1F). Extensive alternative splicing resulting in multiple isoforms has been observed in the mouse ortholog. Similar alternatively spliced transcripts are inferred to occur in human, and additional variants are likely to occur. Functional note: Calcium-dependent cell-adhesion protein. Essential for maintenance of normal retinal and cochlear function. Reported localization: Cell membrane. Single-pass type I. membrane protein. Secreted. Expression/tissue context: Expressed in brain, lung, kidney, spleen and testis. Found also in the inner and outer synaptic layers, and the nerve fiber layer in adult and fetal retinas. Found in the supporting cells, outer sulcus cells and spiral ganglion of fetal cochlea. Expressed in cytotoxic tumor-derived T- and NK-cell lines as well as biopsies of nasal NK/T-cell lymphomas. Not detected in normal or in vitro activated peripheral blood cells, CD4 or CD8 lymphocytes or NK cells. Isoform 3 is expressed in brain, heart, cerebellum and kidney. CD1 isoforms, such as isoform 1, have a limited pattern of expression and is detected in testis, retina and cochlea. CD2 isoforms, such as isoforms 4 and 5, are expressed in heart, kidney, thymus, spleen, testis, retina and cochlea. CD3 isoforms, such as isoform 6, are widely expressed.

Research relevance and current trends

  • Cancer Susceptibility: Researchers commonly examine how PCDH15 (Breast cancer metastasis-suppressor 1) relates to this theme using model systems and orthogonal readouts.
  • Epigenetics and Nuclear Signaling: Researchers commonly examine how PCDH15 (Breast cancer metastasis-suppressor 1) relates to this theme using model systems and orthogonal readouts.
  • Transcription: Researchers commonly examine how PCDH15 (Breast cancer metastasis-suppressor 1) relates to this theme using model systems and orthogonal readouts.

Common research applications

  • Western blotting: compare relative PCDH15 (Breast cancer metastasis-suppressor 1) levels across conditions; band patterns may reflect isoforms and processing.
  • IHC/IHC-F: assess spatial distribution of PCDH15 (Breast cancer metastasis-suppressor 1) across tissue regions and cell types using matched controls.
  • IF/ICC: evaluate subcellular localization and co-localization patterns; signal can depend on fixation/permeabilization and epitope accessibility.
  • Flow cytometry: quantify target-positive populations and shifts in expression; gating strategy and background staining controls are essential.

Notes for experimental interpretation

  • Specificity notes: No cross reactivity with other proteins.
  • Cross-reactivity: No cross-reactivity with other proteins.
  • Isoforms and PTMs: Apparent size and signal patterns can differ across splice isoforms, proteolytic processing, and post-translational modifications.
  • Controls: Include an isotype control (as relevant), no-primary control for imaging, and orthogonal validation such as KD/KO samples when available.

I have a question about product A03591-1, anti-PCDH15 antibody . I was wondering if it would be possible to conjugate this antibody with biotin. I would need it to be without BSA or sodium azide. I am planning on using a buffer exchange of sodium azide with PBS only. Would there be problems for me to conjugate the antibody and store it in -20 degrees in small aliquots?
It is not recommended storing this antibody with PBS buffer only in -20 degrees. If you want to store it in -20 degrees it is best to add some cryoprotectant like glycerol. If you want carrier free A03591-1 anti-PCDH15 antibody , we can provide it to you in a special formula with trehalose and/or glycerol. These molecules will not interfere with conjugation chemistry and provide a good level of protection for the antibody from degradation. Please be sure to specify this in your purchase order.
