Biological background

Protein function (datasheet): Auxiliary protein of DNA polymerase delta and is involved in the control of eukaryotic DNA replication by increasing the polymerase's processibility during elongation of the leading strand. Induces a robust stimulatory effect on the 3'- 5' exonuclease and 3'-phosphodiesterase, but not apurinic- apyrimidinic (AP) endonuclease, APEX2 activities. Has to be loaded onto DNA in order to be able to stimulate APEX2. Plays a key role in DNA damage response (DDR) by being conveniently positioned at the replication fork to coordinate DNA replication with DNA repair and DNA damage tolerance pathways (PubMed:24939902). Acts as a loading platform to recruit DDR proteins that allow completion of DNA replication after DNA damage and promote postreplication repair: Monoubiquitinated PCNA leads to recruitment of translesion (TLS) polymerases, while 'Lys-63'-linked polyubiquitination of PCNA is involved in error-free pathway and employs recombination mechanisms to synthesize across the lesion. .

Cellular localization (datasheet): Nucleus . Colocalizes with CREBBP, EP300 and POLD1 to sites of DNA damage (PubMed:24939902). Forms nuclear foci representing sites of ongoing DNA replication and vary in morphology and number during S phase. Together with APEX2, is redistributed in discrete nuclear foci in presence of oxidative DNA damaging agents. .

Tissue details (datasheet): Ubiquitously expressed. .

Research relevance and current trends

  • Commonly studied in contexts related to Cell Biology,Cell Cycle,Cell Division,Cell Type Markers,Chromatin Binding Proteins,DNA/RNA,DNA/RNA Binding,DNA Synthesis,Epigenetics and Nuclear Signaling,Tags & Cell Markers,Tumor Antigens,Tumor Biomarkers.
  • Supports comparative expression analysis across conditions, genotypes, or treatments when paired with appropriate controls.
  • Useful for confirming target presence and subcellular distribution using orthogonal readouts (e.g., microscopy vs. immunoblotting).

Common research applications

  • Western blot (WB): Compare relative target abundance and apparent size/isoforms across samples; interpret bands in light of expected MW and potential PTMs.
  • Immunohistochemistry (IHC): Assess tissue distribution and cell-type patterns; interpret staining with appropriate negative controls and antigen context.
  • Immunofluorescence / ICC: Visualize subcellular localization and co-localization patterns; consider fixation/permeabilization compatibility and controls.
  • Flow cytometry: Quantify target-positive populations in single-cell suspensions; pair with viability and isotype/FMO controls conceptually.

Notes for experimental interpretation

  • Consider isoforms, post-translational modifications, and processing that can shift apparent molecular weight or localization.
  • Use appropriate positive and negative controls (e.g., KO/KD, blocking peptide, or isotype controls) to support specificity interpretation.

As a monoclonal antibody, this reagent is expected to recognize a defined epitope, which can support consistency across lots when epitope accessibility is preserved.

I was wanting to use your anti-PCNA Rabbit Monoclonal antibody for IF for mouse skeletal muscle on frozen tissues, but I want to know if it has been validated for this particular application. Has this antibody been validated and is this antibody a good choice for mouse skeletal muscle identification?
It shows on the product datasheet, M00125-1 anti-PCNA Rabbit Monoclonal antibody has been tested for Flow Cytometry, IP, IF, IHC, ICC, WB on human, mouse, rat tissues. We have an innovator award program that if you test this antibody and show it works in mouse skeletal muscle in IHC-frozen, you can get your next antibody for free.
We ordered your anti-PCNA Rabbit Monoclonal antibody for IHC on skeletal muscle a few months ago. I am using rat, and We are going to use the antibody for IP next. you antibody examining skeletal muscle as well as fetal brain cortex in our next experiment. Do you have any suggestion on which antibody would work the best for IP?
I have checked the website and datasheets of our anti-PCNA Rabbit Monoclonal antibody and it appears that M00125-1 has been validated on rat in both IHC and IP. Thus M00125-1 should work for your application. Our Boster satisfaction guarantee will cover this product for IP in rat even if the specific tissue type has not been validated. We do have a comprehensive range of products for IP detection and you can check out our website bosterbio.com to find out more information about them.
