Product Overview
This rabbit polyclonal antibody targets 2',5'-phosphodiesterase 12 (gene PDE12) in human, mouse and rat samples and is validated for Western blot, flow cytometry and ELISA. The predicted molecular weight is 67.4 kDa, and the supplier reports an observed band at about 70 kDa.
PDE12 is a mitochondrial 2′,5′-phosphodiesterase that degrades 2′,5′-linked oligoadenylates (2-5A), the second messengers made by interferon-induced OAS enzymes to activate RNase L. By removing 2-5A it acts as a brake on this antiviral pathway, and it is studied in innate immunity and mitochondrial RNA processing.

| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human THP-1 cells, human PC-3 cells, human RT4 cells, human U251 cells, human Caco-2 cells, human HepG2 cells, human HeLa cells, human HEL cells, rat brain tissue, rat thymus tissue, rat PC-12 cells, mouse brain tissue, mouse thymus tissue, mouse NIH/3T3 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.25 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with a confirmed signal (flow cytometry): Caco-2 cells.
Immunogen
Recombinant human PDE12 fragment (Glu36–Lys607), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 89.9% and 91.3%, respectively.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places 2',5'-phosphodiesterase 12 in the mitochondrion matrix. Tissue expression noted by UniProt (human): Ubiquitous. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
