Product Overview
This rabbit polyclonal antibody recognizes myomegalin (gene PDE4DIP) in human samples and is validated for Western blot, IHC, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 265.1 kDa, and the supplier reports an observed band at about 150 kDa.
Myomegalin (PDE4DIP) is a large coiled-coil protein first identified as a partner of PDE4D that anchors cAMP signaling components to the Golgi and centrosome. Certain isoforms work with AKAP9 and CDK5RAP2 to promote microtubule nucleation and to tether non-centrosomal microtubules to the Golgi, supporting polarized cell migration and spindle orientation.

|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Human) |
| Immunocytochemistry / Immunofluorescence | 5 µg/mL (Human) |

Samples with a confirmed band (WB): human HeLa cells, human HEK293 cells, human K562 cells, human U-87MG cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human colorectal cancer tissue, human placenta tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): MCF-7 cells.
Samples with a confirmed signal (flow cytometry): A549 cells.
Immunogen
Recombinant human PDE4DIP fragment (Gln96–Met1254), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human. UniProt places myomegalin in the Golgi apparatus and SUBCELLULAR LOCATION:. Tissue expression noted by UniProt (human): Highly expressed in adult and fetal heart, in skeletal muscle and, to a lower extent, in brain and placenta. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
