Product Overview
This rabbit polyclonal antibody targets high affinity cAMP-specific and IBMX-insensitive 3',5'-cyclic phosphodiesterase 8A (gene PDE8A) in human samples and is validated for Western blot, flow cytometry and ELISA. The predicted molecular weight is 93.3 kDa, and the supplier reports an observed band at about 93 kDa.
PDE8A is a high-affinity, cAMP-specific phosphodiesterase that is insensitive to the broad inhibitor IBMX. It helps set basal cAMP levels, has been linked to steroidogenic and germ cell regulation, and can bind RAF1 to modulate EGF-driven ERK signaling. It is studied in endocrine signaling and T cell biology.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human) |
|---|---|
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human RT4 cells, human A549 cells, human HeLa cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with a confirmed signal (flow cytometry): SH-SY5Y cells.
Immunogen
Recombinant human PDE8A fragment (Arg32–Glu495), expressed in E. coli. Sequence identity with the mouse ortholog: 80.5%.
Reactivity Notes
The supplier lists reactivity with human. Tissue expression noted by UniProt (human): Expressed in most tissues except thymus and peripheral blood leukocytes. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
