Product Overview
This rabbit polyclonal antibody recognizes [Pyruvate dehydrogenase (acetyl-transferring)] kinase isozyme 1, mitochondrial (gene PDK1) in human, mouse and rat samples and is validated for Western blot, flow cytometry and ELISA. The predicted molecular weight is 49.2 kDa, and the supplier reports an observed band at about 46 kDa.
Pyruvate dehydrogenase kinase 1 (PDK1) is a mitochondrial kinase that phosphorylates the PDHA subunit and switches off the pyruvate dehydrogenase complex, reducing conversion of pyruvate to acetyl-CoA. It is a direct HIF-1 target that shifts cells toward glycolysis under hypoxia, and it is studied in cancer metabolism. It should not be confused with PDPK1, the AGC kinase also called PDK1.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human, Mouse, Rat) |
|---|---|
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human 293T cells, rat heart tissue, rat RH35 cells, mouse heart tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with a confirmed signal (flow cytometry): 293T cells.
Immunogen
Recombinant human PDK1 fragment (Tyr114–Ala436), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 94.7% and 95.7%, respectively.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places [Pyruvate dehydrogenase (acetyl-transferring)] kinase isozyme 1, mitochondrial in the mitochondrion matrix. Tissue expression noted by UniProt (human): Expressed predominantly in the heart. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
