Product Overview
This rabbit polyclonal antibody detects [Pyruvate dehydrogenase (acetyl-transferring)] kinase isozyme 4, mitochondrial (gene PDK4) in human and rat samples and is validated for Western blot. The predicted molecular weight is 46.5 kDa, and the supplier reports an observed band at about 46 kDa.
Pyruvate dehydrogenase kinase 4 (PDK4) is a mitochondrial kinase that shuts down the pyruvate dehydrogenase complex by phosphorylating its E1α subunit. Its expression rises strongly in fasting, diabetes and high-fat feeding, conserving glucose by shifting muscle and heart toward fatty acid oxidation, so it is a common marker in studies of metabolic flexibility.

Validated Applications
| Western blot | 0.1–0.5 µg/mL (Human, Rat) |
|---|
Samples with a confirmed band (WB): human HepG2 cells, human HeLa cells, human A549 cells, rat skeletal muscle tissue, rat heart tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Immunogen
Synthetic peptide from the N-terminal region of human PDK4. It differs from the mouse and rat sequences at three residues.
Reactivity Notes
The supplier lists reactivity with human and rat. UniProt places [Pyruvate dehydrogenase (acetyl-transferring)] kinase isozyme 4, mitochondrial in the mitochondrion matrix. Tissue expression noted by UniProt (human): Ubiquitous. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
