Product Overview
This rabbit polyclonal antibody detects pyruvate dehydrogenase phosphatase regulatory subunit, mitochondrial (gene PDPR) in human, mouse and rat samples and is validated for Western blot, flow cytometry and ELISA. The predicted molecular weight is 99.4 kDa, and the supplier reports an observed band at about 99 kDa.
PDPR is the regulatory subunit of mitochondrial pyruvate dehydrogenase phosphatase. It lowers the sensitivity of the catalytic subunit PDP1 to Mg2+, an inhibition that the polyamine spermine can relieve, and so it helps fine-tune reactivation of the pyruvate dehydrogenase complex.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Mouse, Rat) |
|---|---|
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): rat heart tissue, mouse heart tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with a confirmed signal (flow cytometry): Jurkat cells.
Immunogen
Recombinant human PDPR fragment (Glu155–Glu655), expressed in E. coli. Sequence identity with the mouse ortholog: 94.4%.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places pyruvate dehydrogenase phosphatase regulatory subunit, mitochondrial in the mitochondrion matrix. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
