Product Overview
This rabbit polyclonal antibody recognizes pancreas/duodenum homeobox protein 1 (gene PDX1) in human samples and is validated for Western blot, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 30.8 kDa, and the supplier reports an observed band at about 40 kDa.
PDX1 (also IPF1) is a homeodomain transcription factor essential for pancreas development. It specifies early pancreatic progenitors and, in adult β-cells, drives expression of insulin and other genes needed for glucose-regulated insulin secretion, so it is a key marker in β-cell biology, diabetes research and stem cell differentiation.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human) |
|---|---|
| Immunocytochemistry / Immunofluorescence | 5 µg/mL (Human) |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |

Samples with a confirmed band (WB): human PC-3 cells, human HeLa cells, human 293T cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (ICC/IF): HeLa cells.
Samples with a confirmed signal (flow cytometry): Caco-2 cells.
Immunogen
Recombinant human PDX1 fragment (Met1–Glu145), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 87.7% and 86.9%, respectively.
Reactivity Notes
The supplier lists reactivity with human. UniProt places pancreas/duodenum homeobox protein 1 in the nucleus and cytoplasm. Tissue expression noted by UniProt (human): Duodenum and pancreas (Langerhans islet beta cells and small subsets of endocrine non-beta-cells, at low levels in acinar cells). The samples tested by the supplier (listed above) are a practical starting point for positive controls.
