Product Overview

Clone 23P73 is a rabbit monoclonal antibody that recognizes na(+)/H(+) exchange regulatory cofactor NHE-RF3 (gene PDZK1) in human samples and is validated for Western blot and immunoprecipitation. The predicted molecular weight is 57.1 kDa, and the supplier reports an observed band at about 70 kDa.

PDZK1 (NHERF3) is a scaffold protein with four PDZ domains found at the apical membrane of epithelial cells in the kidney, intestine and liver. It binds transporters and receptors such as the HDL receptor SR-BI (SCARB1), CFTR and the anion exchanger SLC26A6, controlling their surface expression, and in the liver it is needed for normal SR-BI levels and cholesterol handling.

Validated Applications

Western blot 1:500–1:2000
Immunoprecipitation 1:50

Samples with a confirmed band (WB): T47-D cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Immunogen

Synthetic peptide based on the sequence of human PDZK1.

Reactivity Notes

The supplier lists reactivity with human. UniProt places na(+)/H(+) exchange regulatory cofactor NHE-RF3 in the membrane and cell membrane. Tissue expression noted by UniProt (human): Expression is limited to epithelial cells. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunoprecipitation 1:50. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 57.1 kDa; observed band at about 70 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human PDZK1.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
