Product Overview
This rabbit polyclonal antibody targets xaa-Pro dipeptidase (gene PEPD) in human, mouse and rat samples and is validated for Western blot, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 54.5 kDa, and the supplier reports an observed band at about 60 kDa.
Prolidase (PEPD) is a cytosolic dipeptidase that cleaves dipeptides carrying proline or hydroxyproline at the C-terminus, such as Gly-Pro. Because collagen is rich in these imino acids, prolidase is important for recycling proline during collagen turnover, and its inherited deficiency causes prolidase deficiency with skin ulcers and other defects.

Samples with a confirmed band (WB): human 293T cells, human SH-SY5Y cells, human HeLa cells, human HepG2 cells, rat small intestine tissue, mouse small intestine tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (ICC/IF): HeLa cells.
Samples with a confirmed signal (flow cytometry): 293T cells.
Immunogen
Recombinant human PEPD fragment (Arg33–Lys493), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 89.4%.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
