Product Overview
This rabbit polyclonal antibody targets thioredoxin-dependent peroxide reductase, mitochondrial (gene PRDX3) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF and flow cytometry. The predicted molecular weight is 27.7 kDa, and the supplier reports an observed band at about 26 kDa.
Peroxiredoxin 3 (PRDX3) is the mitochondrial member of the peroxiredoxin family, reducing hydrogen peroxide generated during respiration with the help of the mitochondrial thioredoxin system. It protects mitochondria from oxidative damage and has been linked to NF-κB regulation, making it relevant to studies of mitochondrial function, metabolism and cell survival.

Validated Applications
| Western blot | 0.1–0.5 µg/mL (Human, Mouse, Rat) |
|---|---|
| IHC (paraffin sections) | 0.5–1 µg/mL (Human, Rat) |
| Immunocytochemistry / Immunofluorescence | 5 µg/mL (Human) |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
Samples with a confirmed band (WB): human LN229 cells, rat C6 cells, human HepG2 cells, human A549 cells, human 293T cells, human Caco-2.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 12% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): rat intestine tissue, human lung cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): A549 cells.
Samples with a confirmed signal (flow cytometry): K562 cells.
Immunogen
Synthetic peptide from the C-terminal region of human peroxiredoxin 3. It differs from the mouse and rat sequences at three residues.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places thioredoxin-dependent peroxide reductase, mitochondrial in the mitochondrion, cytoplasm and early endosome. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
