Overview

This antibody is intended for detection of PF4 (Platelet factor 4) in biological samples using common immunoassay formats. It is typically selected based on target identity, species reactivity, clonality/clone information, and detection modality.

Vendor notes: Boster Bio Anti-PF4 Antibody Picoband® catalog # PB10011. Tested in ELISA, IHC, WB applications. This antibody reacts with Human, Mouse, Rat. The brand Picoband indicates this is a premium antibody that guarantees superior quality, high affinity, and strong signals with minimal background in Western blot applications. Only our best-performing antibodies are designated as Picoband, ensuring unmatched performance.

Key elements and design rationale

  • Antibody format: Rabbit Polyclonal Rabbit IgG
  • Immunogen / epitope context: E. coli-derived human PF4 recombinant protein (Position: E32-S101). Human PF4 shares 75.4% amino acid (aa) sequence identity with both mouse and rat PF4. (reported region: E32-S101).
  • Molecular weight context: reported MW: 11 kDa; calculated MW: 11 kDa
  • Reactivity: Human,Mouse,Rat
  • Applications: ELISA, IHC, WB

As a polyclonal antibody, the reagent recognizes multiple epitopes on the target, which can improve detection robustness but may increase sensitivity to sample-dependent epitope changes.

Biological background

Platelet factor 4; Platelet factor 4. Platelet factor 4 (PF4) is a small cytokine belonging to the CXC chemokine family that is also known as chemokine (C-X-C motif) ligand 4 (CXCL4). By in situ hybridization, the CXCL4 gene is mapped to chromosome 4q12-q21. Its major physiologic role appears to be neutralization of heparin-like molecules on the endothelial surface of blood vessels, thereby inhibiting local antithrombin III activity and promoting coagulation. As a strong chemoattractant for neutrophils and fibroblasts, PF4 probably has a role in inflammation and wound repair. Functional note: Released during platelet aggregation. Neutralizes the anticoagulant effect of heparin because it binds more strongly to heparin than to the chondroitin-4-sulfate chains of the carrier molecule. Chemotactic for neutrophils and monocytes. Inhibits endothelial cell proliferation, the short form is a more potent inhibitor than the longer form. . Reported localization: Secreted. Expression/tissue context: Highly expressed in heart, placenta, pancreas, lung and liver and weakly expressed in spleen, lymph nodes and thymus. .

Research relevance and current trends

  • Atherosclerosis: Researchers commonly examine how PF4 (Platelet factor 4) relates to this theme using model systems and orthogonal readouts.
  • Cardiovascular: Researchers commonly examine how PF4 (Platelet factor 4) relates to this theme using model systems and orthogonal readouts.
  • Chemokines: Researchers commonly examine how PF4 (Platelet factor 4) relates to this theme using model systems and orthogonal readouts.

Common research applications

  • Western blotting: compare relative PF4 (Platelet factor 4) levels across conditions; band patterns may reflect isoforms and processing.
  • IHC/IHC-F: assess spatial distribution of PF4 (Platelet factor 4) across tissue regions and cell types using matched controls.
  • ELISA-compatible use: when applicable, interpret signal as relative abundance across sample sets with consistent handling and dilution strategy.

Notes for experimental interpretation

  • Specificity notes: No cross reactivity with other proteins.
  • Cross-reactivity: No cross-reactivity with other proteins
  • Family / similarity context: Belongs to the transient receptor (TC 1.A.4) family. STrpC subfamily. TRPC4 sub-subfamily.
  • Isoforms and PTMs: Apparent size and signal patterns can differ across splice isoforms, proteolytic processing, and post-translational modifications.
  • Controls: Include an isotype control (as relevant), no-primary control for imaging, and orthogonal validation such as KD/KO samples when available.

Our team were happy with the WB result of your anti-PF4 antibody. However we have observed positive staining in leukocyte secreted. using this antibody. Is that expected? Could you tell me where is PF4 supposed to be expressed?
From literature, leukocyte does express PF4. Generally PF4 expresses in secreted. Regarding which tissues have PF4 expression, here are a few articles citing expression in various tissues: Leukocyte, Pubmed ID: 7644496
We ordered your anti-PF4 antibody for WB on mononuclear cell a few years ago. I am using human, and I plan to use the antibody for ELISA next. We are interested in examining mononuclear cell as well as leukocyte in our next experiment. Could you please give me some suggestion on which antibody would work the best for ELISA?
I took a look at the website and datasheets of our anti-PF4 antibody and I see that PB10011 has been validated on human in both WB and ELISA. Thus PB10011 should work for your application. Our Boster satisfaction guarantee will cover this product for ELISA in human even if the specific tissue type has not been validated. We do have a comprehensive range of products for ELISA detection and you can check out our website bosterbio.com to find out more information about them.
