Product Overview

Clone ADII-16 is a rabbit monoclonal antibody that recognizes 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase 3 (gene PFKFB3) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 59.6 kDa, and the supplier reports an observed band at about 58 kDa.

PFKFB3 is an inducible isoform of 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase with very high kinase activity, so it strongly raises fructose 2,6-bisphosphate and stimulates glycolysis. It is induced by hypoxia and growth signals and is highly expressed in proliferating, cancer and endothelial cells, making it a well-known target in glycolysis and angiogenesis research.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. Tissue expression noted by UniProt (human): Ubiquitous. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunohistochemistry 1:50–1:200; Immunocytochemistry / Immunofluorescence 1:50–1:200; Flow cytometry 1:50; Immunoprecipitation 1:50. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.Has this antibody been tested on monkey samples?
A.Reactivity with monkey samples has not been tested by the supplier. Aligning the immunogen sequence with the monkey ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 59.6 kDa; observed band at about 58 kDa.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human PFKFB3.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
