Product Overview
This rabbit polyclonal antibody detects 6-phosphogluconate dehydrogenase, decarboxylating (gene PGD) in human, mouse and rat samples and is validated for Western blot, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 53.1 kDa, and the supplier reports an observed band at about 48 kDa.
6-phosphogluconate dehydrogenase (PGD) catalyzes the third step of the oxidative pentose phosphate pathway, oxidatively decarboxylating 6-phosphogluconate to yield ribulose 5-phosphate, CO2 and NADPH. It thus supplies NADPH for antioxidant defense and biosynthesis as well as pentose sugars for nucleotides, and it is studied in redox and cancer metabolism.

Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.25 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (ICC/IF): A431 cells.
Samples with a confirmed signal (flow cytometry): HepG2 cells.
Immunogen
Recombinant human PGD fragment (Asp24–Arg447), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 93.9% and 92.9%, respectively.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places 6-phosphogluconate dehydrogenase, decarboxylating in the cytoplasm. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
