Product Overview
This rabbit polyclonal antibody targets 6-phosphogluconolactonase (gene PGLS) in human, mouse and rat samples and is validated for Western blot, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 27.5 kDa, and the supplier reports an observed band at about 28 kDa.
6-phosphogluconolactonase (PGLS) catalyzes the second step of the oxidative pentose phosphate pathway, hydrolyzing 6-phosphogluconolactone to 6-phosphogluconate. This step keeps the pathway flowing toward NADPH and ribose 5-phosphate production, and PGLS is studied in redox metabolism.

Samples with a confirmed band (WB): human Jurkat cells, human Ramos cells, human CCRF-CEM cells, human U251 cells, rat liver tissue, mouse heart tissue, mouse liver tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (ICC/IF): U2OS cells.
Samples with a confirmed signal (flow cytometry): Jurkat cells.
Immunogen
Recombinant human PGLS fragment (Gln16–Leu258), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 88.9% and 88.5%, respectively.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places 6-phosphogluconolactonase in the cytoplasm. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
