Product Overview
This rabbit polyclonal antibody targets phosphoacetylglucosamine mutase (gene PGM3) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 59.9 kDa, and the supplier reports an observed band at about 63 kDa.
PGM3 (phosphoacetylglucosamine mutase) converts N-acetylglucosamine 6-phosphate into N-acetylglucosamine 1-phosphate in the hexosamine pathway that produces UDP-GlcNAc. UDP-GlcNAc is needed for N- and O-linked glycosylation, and PGM3 deficiency causes an inherited immunodeficiency with high IgE, so it is studied in glycosylation and immunity.

Samples with a confirmed band (WB): human A549 cells, human HepG2 cells, human Jurkat cells, human k562 cells, mouse heart tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human testicular seminoma tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): HeLa cells.
Samples with confirmed staining (ICC/IF): rat brain tissue.
Samples with a confirmed signal (flow cytometry): HepG2 cells.
Immunogen
Recombinant human PGM3 fragment (Lys112–Ala531), expressed in E. coli. Sequence identity with the mouse ortholog: 88.1%.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. Tissue expression noted by UniProt (human): Found in many tissues except lung. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
