Key elements and design rationale

  • Target: UCHL1 (Ubiquitin carboxyl-terminal hydrolase isozyme L1).
  • Antibody format: Monoclonal; clone AOCE-21; isotype Rabbit IgG.
  • Host: Rabbit.
  • Species reactivity: Human,Mouse,Rat (confirm in your model system with appropriate controls).

This description is intended to help interpret the antibody design and the biological context of the target using the fields provided in the catalog record, alongside general experimental considerations.

Biological background

UCHL1 (protein: Glycogen synthase kinase-3 beta (gsk3b)) is a commonly studied target in molecular and cellular biology. Functional context (as provided): Ubiquitin-protein hydrolase involved both in the processing of ubiquitin precursors and of ubiquitinated proteins. This enzyme is a thiol protease that recognizes and hydrolyzes a peptide bond at the C-terminal glycine of ubiquitin. Also binds to free monoubiquitin and may prevent its degradation in lysosomes. The homodimer may have ATP-independent ubiquitin ligase activity. . Reported cellular localization context: Cytoplasm . Endoplasmic reticulum membrane ; Lipid-anchor . About 30% of total UCHL1 is associated with membranes in brain. Tissue expression notes (as provided): Found in neuronal cell bodies and processes throughout the neocortex (at protein level). Expressed in neurons and cells of the diffuse neuroendocrine system and their tumors. Weakly expressed in ovary. Down-regulated in brains from Parkinson disease and Alzheimer disease patients. .

Research relevance and current trends

  • Research context keywords from the source record include: Cell Type Marker,Cell Type Markers,Neurodegenerative Disease,Neurology Process,Neuroscience,Tags & Cell Markers.
  • Current studies often focus on connecting target abundance/localization to pathway perturbations across models, tissues, and cell states.
  • Quantitative and multiplexed assays (e.g., imaging + immunoblot panels) are commonly used to compare phenotypes across conditions and time-courses.

Common research applications

  • Western blotting (WB): assess relative target abundance across samples, treatments, or time-points.
  • Immunohistochemistry (IHC): evaluate spatial distribution of target-positive staining in tissue architecture.
  • Immunoprecipitation (IP): enrich target complexes for downstream immunoblot or interaction analyses.

Workflow ideas (metafield): Validate UCHL1 antibody specificity using KO/KD control samples (WB/IF/IHC as appropriate), Detect UCHL1 expression by Western blot in cell or tissue lysates, Detect UCHL1 in FFPE tissue sections by immunohistochemistry, Enrich UCHL1 by immunoprecipitation from lysates for downstream analysis

Notes for experimental interpretation

  • Consider isoforms and post-translational modifications (PTMs) that may shift apparent molecular weight or epitope accessibility.
  • Apparent molecular weight may vary by sample type and processing (observed MW: 22 kDa; calculated MW: 24824 MW).
  • Control concepts: include appropriate negative controls (e.g., isotype, KO/KD samples) and orthogonal validation when feasible.

Additional product details (from the source record)

  • Molecular weight (observed): 22 kDa
  • Cellular localization (provided): Cytoplasm . Endoplasmic reticulum membrane ; Lipid-anchor . About 30% of total UCHL1 is associated with membranes in brain.
  • Tissue details (provided): Found in neuronal cell bodies and processes throughout the neocortex (at protein level). Expressed in neurons and cells of the diffuse neuroendocrine system and their tumors. Weakly expressed in ovary. Down-regulated in brains from Parkinson disease and Alzheimer disease patients. .

Does M01018-1 anti-PGP9.5 Rabbit Monoclonal antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?
You can see on the product datasheet, M01018-1 anti-PGP9.5 Rabbit Monoclonal antibody as been validated on IF. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.
I see that the anti-PGP9.5 Rabbit Monoclonal antibody M01018-1 works with IF, what is the protocol used to produce the result images on the product page?
You can find protocols for IF on the "support/technical resources" section of our navigation menu. If you have any further questions, please send an email to support@bosterbio.com
