Product Overview
This rabbit polyclonal antibody recognizes polyhomeotic-like protein 1 (gene PHC1) in human samples and is validated for Western blot, ICC/IF and ELISA. The predicted molecular weight is 105.5 kDa, and the supplier reports an observed band at about 140 kDa.
PHC1 (polyhomeotic homolog 1) is a subunit of canonical Polycomb repressive complex 1 (PRC1). Through its SAM domain it helps PRC1 form higher-order clusters that compact chromatin, and together with H2AK119 monoubiquitination by PRC1 this keeps developmental genes such as Hox genes stably silenced. It is studied in epigenetics and stem cell biology.

Samples with a confirmed band (WB): human U2OS cells, human SH-SY5Y cells, human Jurkat cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (ICC/IF): U2OS cells.
Immunogen
Recombinant human PHC1 fragment (Arg23–Lys642), expressed in E. coli. Sequence identity with the mouse ortholog: 93.6%.
Reactivity Notes
The supplier lists reactivity with human. UniProt places polyhomeotic-like protein 1 in the nucleus. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
