Product Overview
Clone HEA-1 is a rabbit monoclonal antibody that recognizes RAC-alpha serine/threonine-protein kinase (gene AKT1) phosphorylated at Thr450 in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF and immunoprecipitation. The predicted molecular weight is 55.7 kDa, and the supplier reports an observed band at about 65 kDa.
AKT1 (protein kinase Bα) is one of three AKT serine/threonine kinases activated downstream of PI3K by growth factors and insulin. By phosphorylating many substrates, it promotes cell survival, growth, proliferation, glucose uptake and angiogenesis, and it is one of the most studied nodes in cancer and metabolic signaling.

Samples with a confirmed band (WB): human HeLa cells, human MCF-7 cells, rat brain tissue, rat PC-12 cells, mouse brain tissue, mouse NIH/3T3 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:1000 overnight at 4 °C, HRP-conjugated secondary antibody at 1:1000, ECL detection.
Samples with confirmed staining (IHC): human colorectal adenocarcinoma tissue, human prostate cancer tissue, human lung adenocarcinoma tissue, intestinal diffuse large B-cells lymphoma tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): A549 cells.
Immunogen
Synthetic peptide based on the sequence of human AKT1.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places RAC-alpha serine/threonine-protein kinase in the cytoplasm, nucleus, cell membrane and mitochondrion intermembrane space. Tissue expression noted by UniProt (human): Expressed in prostate cancer and levels increase from the normal to the malignant state (at protein level). The samples tested by the supplier (listed above) are a practical starting point for positive controls. Because the antibody is phospho-specific, signal depends on the phosphorylation state of the sample; a phosphatase-treated lysate is a useful negative control.
