Product Overview

Clone AEBH-2 is a rabbit monoclonal antibody that recognizes bcl2-associated agonist of cell death (gene BAD) phosphorylated at Ser112 in human, mouse and rat samples and is validated for Western blot and immunoprecipitation. The predicted molecular weight is 18.4 kDa, and the supplier reports an observed band at about 23 kDa.

BAD is a pro-apoptotic BH3-only member of the Bcl-2 family. When dephosphorylated it binds Bcl-2 and Bcl-xL and frees BAX and BAK to trigger apoptosis, while survival signals such as AKT phosphorylate BAD and sequester it with 14-3-3 proteins. It is widely used to study growth factor-dependent cell survival.

Validated Applications

Western blot 1:500–1:2000
Immunoprecipitation 1:20

Samples with a confirmed band (WB): HeLa cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Immunogen

Synthetic peptide based on the sequence of human bad Promotes cell death.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places bcl2-associated agonist of cell death in the mitochondrion outer membrane and cytoplasm. Tissue expression noted by UniProt (human): Expressed in a wide variety of tissues. The samples tested by the supplier (listed above) are a practical starting point for positive controls. Because the antibody is phospho-specific, signal depends on the phosphorylation state of the sample; a phosphatase-treated lysate is a useful negative control.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunoprecipitation 1:20. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.Is a carrier-free (BSA-free) version available, for example for conjugation?
A.Yes. A carrier-free formulation without BSA, stabilized with trehalose and/or glycerol, can be prepared on request and is compatible with common conjugation chemistries. Please note this when ordering or contact info@biohippo.com. For storage at −20 °C, keep a cryoprotectant such as glycerol in the buffer rather than PBS alone.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 18.4 kDa; observed band at about 23 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human bad Promotes cell death.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
