Product Overview

Clone 32B03 is a rabbit monoclonal antibody that recognizes breast cancer anti-estrogen resistance protein 1 (gene BCAR1) phosphorylated at Tyr410 in human samples and is validated for Western blot. The predicted molecular weight is 93.4 kDa, and the supplier reports an observed band at about 130 kDa.

BCAR1 (p130Cas) is a docking protein at focal adhesions that becomes tyrosine phosphorylated by FAK and SRC after integrin engagement. Its phosphorylated substrate domain recruits CRK and other adaptors that drive actin remodeling and cell migration. BCAR1 was identified through resistance to antiestrogens in breast cancer cells and is widely studied in adhesion signaling.

Validated Applications

Western blot 1:500–1:2000

Samples with a confirmed band (WB): HeLa cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Immunogen

Synthetic phosphopeptide around Tyr410 of human BCAR1.

Reactivity Notes

The supplier lists reactivity with human. UniProt places breast cancer anti-estrogen resistance protein 1 in the cell junction, cytoplasm and cell projection. Tissue expression noted by UniProt (human): Expressed in B-cells (at protein level). The samples tested by the supplier (listed above) are a practical starting point for positive controls. Because the antibody is phospho-specific, signal depends on the phosphorylation state of the sample; a phosphatase-treated lysate is a useful negative control.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 93.4 kDa; observed band at about 130 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic phosphopeptide around Tyr410 of human BCAR1.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
