Product Overview
Clone FII-2 is a rabbit monoclonal antibody that recognizes tyrosine-protein kinase BTK phosphorylated at Tyr551 in human samples and is validated for Western blot and ICC/IF. The predicted molecular weight is 76.3 kDa, and the supplier reports an observed band at about 74 kDa.
Bruton′s tyrosine kinase (BTK) is a TEC family kinase that relays B-cell receptor signals to PLCγ2, calcium release and NF-κB activation, and it also acts downstream of Fc and Toll-like receptors in myeloid cells. BTK is essential for B-cell development, its mutations cause X-linked agammaglobulinemia, and it is the target of inhibitors such as ibrutinib.

Reactivity Notes
The supplier lists reactivity with human. UniProt places tyrosine-protein kinase BTK in the cytoplasm, cell membrane, nucleus and membrane raft. Tissue expression noted by UniProt (human): Predominantly expressed in B-lymphocytes. The samples tested by the supplier (listed above) are a practical starting point for positive controls. Because the antibody is phospho-specific, signal depends on the phosphorylation state of the sample; a phosphatase-treated lysate is a useful negative control.
