Product Overview
Clone EAC-13 is a rabbit monoclonal antibody that recognizes myc proto-oncogene protein phosphorylated at Ser62 in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF and immunoprecipitation. The predicted molecular weight is 48.8 kDa, and the supplier reports an observed band at about 57 kDa.
c-Myc (MYC) is a bHLH-leucine zipper transcription factor that pairs with MAX, binds E-box sequences and broadly amplifies the transcription of genes for growth, ribosome biogenesis, metabolism and proliferation. It also supports stem cell self-renewal and reprogramming. MYC is one of the most frequently deregulated oncogenes in human cancer.

Samples with a confirmed band (WB): human HeLa cells, human K562 cells, human MCF-7 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human lung carcinoma tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Immunogen
Synthetic phosphopeptide around Ser62 of human c-Myc.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places myc proto-oncogene protein in the nucleus, cytoplasm and chromosome. The samples tested by the supplier (listed above) are a practical starting point for positive controls. Because the antibody is phospho-specific, signal depends on the phosphorylation state of the sample; a phosphatase-treated lysate is a useful negative control.
