Product Overview
Clone GHC-3 is a rabbit monoclonal antibody that recognizes G1/S-specific cyclin-E1 (gene CCNE1) phosphorylated at Thr77 in human and mouse samples and is validated for Western blot, IHC and ICC/IF. The predicted molecular weight is 47.1 kDa, and the supplier reports an observed band at about 47 kDa.
Cyclin E1 (CCNE1) activates CDK2 in late G1 to promote the transition into S phase and the start of DNA replication. It is tightly regulated by ubiquitin-mediated degradation, and CCNE1 amplification is well documented in several cancers, notably high-grade serous ovarian cancer.

Samples with a confirmed band (WB): human HeLa cells, human K562 cells, human Caco-2 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human testis tissue, human placenta tissue, mouse testis tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Synthetic peptide based on the sequence of human cyclin E1.
Reactivity Notes
The supplier lists reactivity with human and mouse. UniProt places G1/S-specific cyclin-E1 in the nucleus. Tissue expression noted by UniProt (human): Highly expressed in testis and placenta. The samples tested by the supplier (listed above) are a practical starting point for positive controls. Because the antibody is phospho-specific, signal depends on the phosphorylation state of the sample; a phosphatase-treated lysate is a useful negative control.
