Product Overview

Clone 31E45 is a rabbit monoclonal antibody that recognizes epidermal growth factor receptor (gene EGFR) phosphorylated at Tyr845 in human samples and is validated for Western blot and ICC/IF. The predicted molecular weight is 134.3 kDa, and the supplier reports an observed band at about 220 kDa.

Epidermal growth factor receptor (EGFR, also called ErbB1 or HER1) is a receptor tyrosine kinase activated by EGF, TGF-α and related ligands. Ligand binding triggers receptor dimerization and activation of RAS/MAPK, PI3K/AKT and other pathways that drive proliferation and survival, and EGFR is a major target in cancer research.

Validated Applications

Western blot 1:500–1:2000
Immunocytochemistry / Immunofluorescence 1:50–1:200

Samples with a confirmed band (WB): A431 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Immunogen

Synthetic phosphopeptide around Tyr845 of human EGFR.

Reactivity Notes

The supplier lists reactivity with human. UniProt places epidermal growth factor receptor in the cell membrane, endoplasmic reticulum membrane, Golgi apparatus membrane and nuclear membrane. Tissue expression noted by UniProt (human): Ubiquitously expressed. The samples tested by the supplier (listed above) are a practical starting point for positive controls. Because the antibody is phospho-specific, signal depends on the phosphorylation state of the sample; a phosphatase-treated lysate is a useful negative control.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunocytochemistry / Immunofluorescence 1:50–1:200. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 134.3 kDa; observed band at about 220 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic phosphopeptide around Tyr845 of human EGFR.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
