Product Overview

Clone 32E48 is a rabbit monoclonal antibody that recognizes eukaryotic translation initiation factor 4E-binding protein 1 (gene EIF4EBP1) phosphorylated at Thr70 in human samples and is validated for Western blot. The predicted molecular weight is 12.6 kDa, and the supplier reports an observed band at about 17 kDa.

4E-BP1 (EIF4EBP1) is a translational repressor that binds eIF4E and prevents it from joining eIF4G. When mTORC1 phosphorylates 4E-BP1, it releases eIF4E and cap-dependent translation increases, so phospho-4E-BP1 is a standard readout of mTOR activity.

Validated Applications

Western blot 1:500–1:2000

Samples with a confirmed band (WB): human SH-SY5Y cells, human HEL cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 12% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Immunogen

Synthetic phosphopeptide around Thr70 of human eIF4EBP1.

Reactivity Notes

The supplier lists reactivity with human. UniProt places eukaryotic translation initiation factor 4E-binding protein 1 in the cytoplasm and nucleus. The samples tested by the supplier (listed above) are a practical starting point for positive controls. Because the antibody is phospho-specific, signal depends on the phosphorylation state of the sample; a phosphatase-treated lysate is a useful negative control.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 12.6 kDa; observed band at about 17 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic phosphopeptide around Thr70 of human eIF4EBP1.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
