Clone 27E43 is a rabbit monoclonal antibody that recognizes receptor tyrosine-protein kinase erbB-2 (gene ERBB2) phosphorylated at Tyr1139 in human samples and is validated for Western blot and immunoprecipitation. The predicted molecular weight is 137.9 kDa, and the supplier reports an observed band at about 185 kDa.
HER2 (ERBB2) is a receptor tyrosine kinase of the EGFR family that has no known ligand of its own but is the preferred partner for other ErbB receptors. Heterodimers containing HER2 signal strongly through MAPK and PI3K-AKT pathways. HER2 amplification drives a subset of breast and gastric cancers, making it one of the most studied oncology targets.

Reactivity Notes
The supplier lists reactivity with human. UniProt places receptor tyrosine-protein kinase erbB-2 in the cell membrane and cell projection. Tissue expression noted by UniProt (human): Expressed in a variety of tumor tissues including primary breast tumors and tumors from small bowel, esophagus, kidney and mouth. The samples tested by the supplier (listed above) are a practical starting point for positive controls. Because the antibody is phospho-specific, signal depends on the phosphorylation state of the sample; a phosphatase-treated lysate is a useful negative control.
