Cellular localization Nucleus. Chromosome, centromere, kinetochore. Cytoplasm, cytoskeleton, microtubule organizing center, centrosome. Cytoplasm, cytoskeleton, spindle. Midbody. During early stages of mitosis, the phosphorylated form is detected on centrosomes and kinetochores. Localizes to the outer kinetochore. Presence of SGOL1 and interaction with the phosphorylated form of BUB1 is required for the kinetochore localization. Localizes onto the central spindle by phosphorylating and docking at midzone proteins KIF20A/MKLP2 and PRC1. Colocalizes with FRY to separating centrosomes and spindle poles from prophase to metaphase in mitosis, but not in other stages of the cell cycle.

Product Overview

Clone 27F40 is a rabbit monoclonal antibody that recognizes fibroblast growth factor receptor 3 (gene FGFR3) phosphorylated at Tyr724 in human and mouse samples and is validated for Western blot, ICC/IF, flow cytometry and immunoprecipitation. The predicted molecular weight is 87.7 kDa, and the supplier reports an observed band at about 100 kDa.

Fibroblast growth factor receptor 3 (FGFR3) is a receptor tyrosine kinase that responds to FGFs and acts as a brake on chondrocyte proliferation in growing bone. Activating FGFR3 mutations cause achondroplasia and related skeletal dysplasias, and FGFR3 alterations are frequent in bladder cancer, so it is studied in bone growth and oncology.

Target Fibroblast growth factor receptor 3 (gene FGFR3; UniProt P22607, human) — phosphorylated at Tyr724

Immunoprecipitation 1:50
Flow cytometry 1:50

Samples with a confirmed band (WB): MCF-7.

Reactivity Notes

The supplier lists reactivity with human and mouse. Tissue expression noted by UniProt (human): Expressed in brain, kidney and testis. The samples tested by the supplier (listed above) are a practical starting point for positive controls. Because the antibody is phospho-specific, signal depends on the phosphorylation state of the sample; a phosphatase-treated lysate is a useful negative control.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunocytochemistry / Immunofluorescence 1:50–1:200; Flow cytometry 1:50; Immunoprecipitation 1:50. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 87.7 kDa; observed band at about 100 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic phosphopeptide around Tyr724 of human FGFR3.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
