Product Overview
Clone BHI-8 is a rabbit monoclonal antibody that recognizes heat shock factor protein 1 (gene HSF1) phosphorylated at Ser326 in human samples and is validated for Western blot, IHC, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 57.3 kDa, and the supplier reports an observed band at about 80 kDa.
Heat shock factor 1 (HSF1) is the master transcription factor of the heat shock response. Under proteotoxic stress it forms trimers, binds heat shock elements and switches on genes for chaperones such as HSP70 and HSP90, which protect cells from misfolded proteins. HSF1 is studied in protein homeostasis, aging, neurodegeneration and cancer.

Samples with a confirmed band (WB): human HeLa cells, human K562 cells, human HepG2 cells, human MCF-7 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:1000, ECL detection.
Samples with confirmed staining (IHC): human breast cancer tissue, human spleen tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Synthetic phosphopeptide around Ser326 of human HSF1.
Reactivity Notes
The supplier lists reactivity with human. UniProt places heat shock factor protein 1 in the nucleus, cytoplasm and chromosome. The samples tested by the supplier (listed above) are a practical starting point for positive controls. Because the antibody is phospho-specific, signal depends on the phosphorylation state of the sample; a phosphatase-treated lysate is a useful negative control.
