Product Overview
Clone 31J16 is a rabbit monoclonal antibody that recognizes tyrosine-protein kinase JAK1 phosphorylated at Tyr1034 in human samples and is validated for Western blot and IHC. The predicted molecular weight is 133.3 kDa, and the supplier reports an observed band at about 133 kDa.
JAK1 (Janus kinase 1) is a cytoplasmic tyrosine kinase bound to many cytokine receptors, including those for interferons, IL-6 family cytokines and cytokines that use the common gamma chain. Upon cytokine binding, JAK1 phosphorylates the receptor and STAT proteins, which then regulate gene expression, and it is a major target of JAK inhibitor research.

Reactivity Notes
The supplier lists reactivity with human. UniProt places tyrosine-protein kinase JAK1 in the endomembrane system. Tissue expression noted by UniProt (human): Expressed at higher levels in primary colon tumors than in normal colon tissue. The samples tested by the supplier (listed above) are a practical starting point for positive controls. Because the antibody is phospho-specific, signal depends on the phosphorylation state of the sample; a phosphatase-treated lysate is a useful negative control.
