Product Overview

Clone 31L42 is a rabbit monoclonal antibody that recognizes low-density lipoprotein receptor-related protein 6 (gene LRP6) phosphorylated at Ser1490 in human samples and is validated for Western blot. The predicted molecular weight is 180.4 kDa, and the supplier reports an observed band at about 180–210 kDa.

LRP6 is a cell-surface coreceptor for canonical Wnt/β-catenin signaling that works together with Frizzled receptors. When Wnt binds, LRP6 is phosphorylated and helps gather Dishevelled and the Axin/GSK3 complex into membrane signalosomes, which blocks β-catenin destruction; it is widely studied in embryonic patterning, bone formation and cancer signaling.

Validated Applications

Western blot 1:500–1:2000

Samples with a confirmed band (WB): HeLa cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Immunogen

Synthetic phosphopeptide around Ser1490 of human LRP6.

Reactivity Notes

The supplier lists reactivity with human. UniProt places low-density lipoprotein receptor-related protein 6 in the cell membrane, endoplasmic reticulum and membrane raft. Tissue expression noted by UniProt (human): Widely coexpressed with LRP5 during embryogenesis and in adult tissues. The samples tested by the supplier (listed above) are a practical starting point for positive controls. Because the antibody is phospho-specific, signal depends on the phosphorylation state of the sample; a phosphatase-treated lysate is a useful negative control.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 180.4 kDa; observed bands at about 180 and 210 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic phosphopeptide around Ser1490 of human LRP6.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
