Cellular localization Cell membrane. Nucleus. Cytoplasm. Cytoplasm, perinuclear region. Golgi apparatus. Membrane ; Lipid-anchor . Accumulates in the nucleus by inhibition of CRM1-mediated nuclear export. Nuclear accumulation is increased by inhibition of its kinase activity. The trafficking from the Golgi apparatus to the plasma membrane occurs in a kinase domain-dependent but kinase activity independent manner and is mediated by exocytic vesicular transport. Detected on plasma membrane lipid rafts.

Product Overview

Clone IFI-12 is a rabbit monoclonal antibody that recognizes tyrosine-protein kinase Lyn phosphorylated at Tyr396 in human and mouse samples and is validated for Western blot, ICC/IF and flow cytometry. The predicted molecular weight is 58.6 kDa, and the supplier reports an observed band at about 56 kDa.

Lyn is a Src-family non-receptor tyrosine kinase expressed mainly in B cells and myeloid cells. It phosphorylates signaling motifs of the B-cell receptor, Fc receptors and other immune receptors, both starting and shutting down their signals; because loss of Lyn in mice leads to autoimmunity, it is studied in B-cell tolerance, allergy and hematopoietic cancers.

Target Tyrosine-protein kinase Lyn (gene LYN; UniProt P07948, human) — phosphorylated at Tyr396

Immunocytochemistry / Immunofluorescence 1:50–1:200
Flow cytometry 1:20

Recommended loading (WB): 20–40 µg of total protein per lane.

Immunogen

Synthetic phosphopeptide around Tyr396 of human lyn.

Reactivity Notes

The supplier lists reactivity with human and mouse. UniProt places tyrosine-protein kinase Lyn in the cell membrane, nucleus, cytoplasm and Golgi apparatus. Tissue expression noted by UniProt (human): Detected in monocytes (at protein level). The samples tested by the supplier (listed above) are a practical starting point for positive controls. Because the antibody is phospho-specific, signal depends on the phosphorylation state of the sample; a phosphatase-treated lysate is a useful negative control.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunocytochemistry / Immunofluorescence 1:50–1:200; Flow cytometry 1:20. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse. Other species are not listed by the supplier.
Q.Is a blocking reagent available for this antibody?
A.A matching blocking reagent for competition (specificity) controls can be supplied on request. Contact info@biohippo.com to request it.
Q.Is a carrier-free (BSA-free) version available, for example for conjugation?
A.Yes. A carrier-free formulation without BSA, stabilized with trehalose and/or glycerol, can be prepared on request and is compatible with common conjugation chemistries. Please note this when ordering or contact info@biohippo.com. For storage at −20 °C, keep a cryoprotectant such as glycerol in the buffer rather than PBS alone.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 58.6 kDa; observed band at about 56 kDa.
Q.What immunogen was used to raise this antibody?
A.Synthetic phosphopeptide around Tyr396 of human lyn.
