Product Overview
Clone 28M76 is a rabbit monoclonal antibody that recognizes DNA replication licensing factor MCM2 phosphorylated at Ser108 in human, mouse and rat samples and is validated for Western blot, ICC/IF and immunoprecipitation. The predicted molecular weight is 101.9 kDa, and the supplier reports an observed band at about 125 kDa.
MCM2 is a subunit of the MCM2-7 complex, the ring-shaped replicative helicase that is loaded onto origins of DNA replication and unwinds DNA during S phase. MCM2 also binds histones and helps handle them at the replication fork. Because MCM proteins are present in proliferating but not resting cells, MCM2 is widely used as a proliferation marker.

Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places DNA replication licensing factor MCM2 in the nucleus and chromosome. The samples tested by the supplier (listed above) are a practical starting point for positive controls. Because the antibody is phospho-specific, signal depends on the phosphorylation state of the sample; a phosphatase-treated lysate is a useful negative control.
