Product Overview
Clone HBO-13 is a rabbit monoclonal antibody that recognizes dual specificity mitogen-activated protein kinase kinase 1 (gene MAP2K1) phosphorylated at Ser298 in human, mouse and rat samples and is validated for Western blot, IHC and ICC/IF. The predicted molecular weight is 43.4 kDa, and the supplier reports an observed band at about 54 kDa.
MEK1 (MAP2K1) is a dual-specificity kinase in the RAS–RAF–MEK–ERK pathway. Activated by RAF, it phosphorylates ERK1 and ERK2 on both a threonine and a tyrosine in their activation loop, turning on the main mitogenic MAPK cascade, and it is a major target in cancer research and of clinically used MEK inhibitors.

Samples with a confirmed band (WB): human HeLa cells, human A431 cells, human U251 cells, human SIHA cells, rat lung tissue, mouse lung tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:1000 overnight at 4 °C, HRP-conjugated secondary antibody at 1:500, ECL detection.
Immunogen
Synthetic peptide based on the sequence of human MEK1.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places dual specificity mitogen-activated protein kinase kinase 1 in the cytoplasm, nucleus and membrane. Tissue expression noted by UniProt (human): Widely expressed, with extremely low levels in brain. The samples tested by the supplier (listed above) are a practical starting point for positive controls. Because the antibody is phospho-specific, signal depends on the phosphorylation state of the sample; a phosphatase-treated lysate is a useful negative control.
