Product Overview

Clone HCG-13 is a rabbit monoclonal antibody that recognizes dual specificity mitogen-activated protein kinase kinase 1 (gene MAP2K1) phosphorylated at Thr292 in human samples and is validated for Western blot. The predicted molecular weight is 43.4 kDa, and the supplier reports an observed band at about 45 kDa.

MEK1 (MAP2K1) is a dual-specificity kinase in the RAS–RAF–MEK–ERK pathway. Activated by RAF, it phosphorylates ERK1 and ERK2 on both a threonine and a tyrosine in their activation loop, turning on the main mitogenic MAPK cascade, and it is a major target in cancer research and of clinically used MEK inhibitors.

Validated Applications

Western blot 1:500–1:2000

Samples with a confirmed band (WB): HeLa cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Immunogen

Synthetic peptide based on the sequence of human MEK1.

Reactivity Notes

The supplier lists reactivity with human. UniProt places dual specificity mitogen-activated protein kinase kinase 1 in the cytoplasm, nucleus and membrane. Tissue expression noted by UniProt (human): Widely expressed, with extremely low levels in brain. The samples tested by the supplier (listed above) are a practical starting point for positive controls. Because the antibody is phospho-specific, signal depends on the phosphorylation state of the sample; a phosphatase-treated lysate is a useful negative control.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
