Product Overview
Clone BDO-20 is a rabbit monoclonal antibody that recognizes cellular tumor antigen p53 (gene TP53) phosphorylated at Ser392 in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF and immunoprecipitation. The predicted molecular weight is 43.7 kDa, and the supplier reports an observed band at about 53 kDa.
p53 (TP53) is a tumor suppressor transcription factor that responds to DNA damage and other stresses by activating genes for cell cycle arrest, DNA repair, senescence or apoptosis. It is mutated in about half of human cancers, making it one of the most studied proteins in cancer biology, with rat models widely used alongside human cells.

Samples with a confirmed band (WB): human 293T cells, human HeLa cells, rat PC-12 cells, mouse RAW264.7 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:500, ECL detection.
Samples with confirmed staining (IHC): mouse colon tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): HeLa cells.
Immunogen
Synthetic phosphopeptide around Ser392 of human p53.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places cellular tumor antigen p53 in the cytoplasm, nucleus, endoplasmic reticulum and mitochondrion matrix. Tissue expression noted by UniProt (human): Ubiquitous. The samples tested by the supplier (listed above) are a practical starting point for positive controls. Because the antibody is phospho-specific, signal depends on the phosphorylation state of the sample; a phosphatase-treated lysate is a useful negative control.
