Product Overview

Clone ADO-20 is a rabbit monoclonal antibody that recognizes cellular tumor antigen p53 (gene TP53) phosphorylated at Ser9 in human samples and is validated for Western blot. The predicted molecular weight is 43.7 kDa, and the supplier reports an observed band at about 53 kDa.

p53 (TP53) is a tumor suppressor transcription factor that responds to DNA damage and other stresses by activating genes for cell cycle arrest, DNA repair, senescence or apoptosis. It is mutated in about half of human cancers, making it one of the most studied proteins in cancer biology, with rat models widely used alongside human cells.

Validated Applications

Western blot 1:500–1:2000

Samples with a confirmed band (WB): HepG2 cells, HeLa cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Immunogen

Synthetic phosphopeptide around Ser9 of human p53.

Reactivity Notes

The supplier lists reactivity with human. UniProt places cellular tumor antigen p53 in the cytoplasm, nucleus, endoplasmic reticulum and mitochondrion matrix. Tissue expression noted by UniProt (human): Ubiquitous. The samples tested by the supplier (listed above) are a practical starting point for positive controls. Because the antibody is phospho-specific, signal depends on the phosphorylation state of the sample; a phosphatase-treated lysate is a useful negative control.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.Has this antibody been tested on monkey samples?
A.Reactivity with monkey samples has not been tested by the supplier. Aligning the immunogen sequence with the monkey ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 43.7 kDa; observed band at about 53 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic phosphopeptide around Ser9 of human p53.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
