Product Overview

Clone BIC-5 is a rabbit monoclonal antibody that recognizes interferon-induced, double-stranded RNA-activated protein kinase (gene EIF2AK2) phosphorylated at Thr446 in human samples and is validated for Western blot, IHC and immunoprecipitation. The predicted molecular weight is 62.1 kDa, and the supplier reports an observed band at about 68 kDa.

PKR (EIF2AK2) is an interferon-induced kinase activated by double-stranded RNA. It phosphorylates eIF2α to shut down protein synthesis and limit viral replication, and it is studied in antiviral immunity, inflammation and the integrated stress response.

Validated Applications

Western blot 1:500–1:2000
Immunohistochemistry 1:50–1:200
Immunoprecipitation 1:50

Samples with confirmed staining (IHC): human colon tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Immunogen

Synthetic phosphopeptide around Thr446 of human PKR.

Reactivity Notes

The supplier lists reactivity with human. UniProt places interferon-induced, double-stranded RNA-activated protein kinase in the cytoplasm and nucleus. Tissue expression noted by UniProt (human): Highly expressed in thymus, spleen and bone marrow compared to non-hematopoietic tissues such as small intestine, liver, or kidney tissues. The samples tested by the supplier (listed above) are a practical starting point for positive controls. Because the antibody is phospho-specific, signal depends on the phosphorylation state of the sample; a phosphatase-treated lysate is a useful negative control.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunohistochemistry 1:50–1:200; Immunoprecipitation 1:50. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 62.1 kDa; observed band at about 68 kDa.
Q.What immunogen was used to raise this antibody?
A.Synthetic phosphopeptide around Thr446 of human PKR.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
