Cellular localization Cytoplasm. Cell membrane. Mitochondrion. Nucleus. Colocalizes with RGS14 and BRAF in both the cytoplasm and membranes. Phosphorylation at Ser-259 impairs its membrane accumulation. Recruited to the cell membrane by the active Ras protein. Phosphorylation at Ser-338 and Ser-339 by PAK1 is required for its mitochondrial localization. Retinoic acid- induced Ser-621 phosphorylated form of RAF1 is predominantly localized at the nucleus.

Product Overview

Clone ICO-18 is a rabbit monoclonal antibody that recognizes RAF proto-oncogene serine/threonine-protein kinase (gene RAF1) phosphorylated at Ser43 in human, mouse and rat samples and is validated for Western blot. The predicted molecular weight is 73.1 kDa, and the supplier reports an observed band at about 73 kDa.

RAF1 (c-Raf) is a serine/threonine kinase that passes signals from active Ras to MEK1/2 and then ERK1/2, a cascade that shapes proliferation, differentiation and survival. Its activity is controlled by phosphorylation at several regulatory residues, and it is studied widely in growth-factor signaling and in Ras-driven cancers.

Validated Applications

Western blot 1:500–1:2000

Samples with a confirmed band (WB): human HeLa cells, rat PC-12 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Immunogen

Synthetic phosphopeptide around Ser43 of human raf1.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places RAF proto-oncogene serine/threonine-protein kinase in the cytoplasm, cell membrane, mitochondrion and nucleus. Tissue expression noted by UniProt (human): In skeletal muscle, isoform 1 is more abundant than isoform 2. The samples tested by the supplier (listed above) are a practical starting point for positive controls. Because the antibody is phospho-specific, signal depends on the phosphorylation state of the sample; a phosphatase-treated lysate is a useful negative control.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
