Product Overview

Clone 31R36 is a rabbit monoclonal antibody that recognizes receptor-interacting serine/threonine-protein kinase 3 (gene RIPK3) phosphorylated at Ser232 in mouse and rat samples and is validated for Western blot. The predicted molecular weight is 56.9 kDa, and the supplier reports an observed band at about 50 kDa.

RIPK3 is a serine/threonine kinase that drives necroptosis, a regulated form of necrotic cell death. Activated through RIPK1, ZBP1 or TRIF, it forms the necrosome and phosphorylates the pseudokinase MLKL, which then damages the plasma membrane. RIPK3 is studied in inflammation, host defense against viruses and tissue injury.

Validated Applications

Western blot 1:500–1:2000

Samples with a confirmed band (WB): rat spleen tissue, rat liver tissue, rat heart tissue, rat L6 cells, mouse spleen tissue, mouse liver tissue, mouse heart tissue, mouse L929 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Immunogen

Synthetic phosphopeptide around Ser232 of human RIP3.

Reactivity Notes

The supplier lists reactivity with mouse and rat. UniProt places receptor-interacting serine/threonine-protein kinase 3 in the cytoplasm and nucleus. Tissue expression noted by UniProt (human): Highly expressed in the pancreas. The samples tested by the supplier (listed above) are a practical starting point for positive controls. Because the antibody is phospho-specific, signal depends on the phosphorylation state of the sample; a phosphatase-treated lysate is a useful negative control.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Mouse, Rat. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 56.9 kDa; observed band at about 50 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic phosphopeptide around Ser232 of human RIP3.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
