Product Overview

Clone HBC-19 is a rabbit monoclonal antibody that recognizes SMAD family member 2 (gene SMAD2) phosphorylated at Ser250 in human, mouse and rat samples and is validated for Western blot and flow cytometry. The predicted molecular weight is 52.3 kDa, and the supplier reports an observed band at about 58 kDa.

SMAD2 is a receptor-regulated SMAD that carries signals from TGF-β and activin receptors. After phosphorylation it forms a complex with SMAD4 that enters the nucleus to control gene transcription, so phospho-SMAD2 is a standard readout of TGF-β pathway activity in development, fibrosis and cancer.

Validated Applications

Western blot 1:500–1:2000
Flow cytometry 1:20

Samples with a confirmed band (WB): human HeLa cells, human HepG2 cells, human HT-1080 cells, rat heart tissue, rat skeletal muscle tissue, mouse heart tissue, mouse skeletal muscle tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:1000 overnight at 4 °C, HRP-conjugated secondary antibody at 1:1000, ECL detection.

Immunogen

Synthetic peptide based on the sequence of human smad2.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places SMAD family member 2 in the cytoplasm and nucleus. Tissue expression noted by UniProt (human): Expressed at high levels in skeletal muscle, endothelial cells, heart and placenta. The samples tested by the supplier (listed above) are a practical starting point for positive controls. Because the antibody is phospho-specific, signal depends on the phosphorylation state of the sample; a phosphatase-treated lysate is a useful negative control.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Flow cytometry 1:20. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 52.3 kDa; observed band at about 58 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human smad2.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
