    Biological background

Protein function (datasheet): Signal transducer and transcription activator that mediates cellular responses to interleukins, KITLG/SCF and other growth factors. May mediate cellular responses to activated FGFR1, FGFR2, FGFR3 and FGFR4. Binds to the interleukin-6 (IL-6)- responsive elements identified in the promoters of various acute- phase protein genes. Activated by IL31 through IL31RA. Cytoplasmic STAT3 represses macroautophagy by inhibiting EIF2AK2/PKR activity. Plays an important role in host defense in methicillin-resistant S.aureus lung infection by regulating the expression of the antimicrobial lectin REG3G (By similarity). .

Cellular localization (datasheet): Cytoplasm. Nucleus. Shuttles between the nucleus and the cytoplasm. Translocated into the nucleus upon tyrosine phosphorylation and dimerization, in response to signaling by activated FGFR1, FGFR2, FGFR3 or FGFR4. Constitutive nuclear presence is independent of tyrosine phosphorylation. Predominantly present in the cytoplasm without stimuli. Upon leukemia inhibitory factor (LIF) stimulation, accumulates in the nucleus. The complex composed of BART and ARL2 plays an important role in the nuclear translocation and retention of STAT3. Identified in a complex with LYN and PAG1.

Tissue details (datasheet): Heart, brain, placenta, lung, liver, skeletal muscle, kidney and pancreas.

    Research relevance and current trends

    • Commonly studied in contexts related to Embryogenesis,Embryonic Stem Cells,Epigenetics and Nuclear Signaling,Hypertrophy,Intracellular,Nuclear Signaling,Nuclear Signaling Pathways,Signal Transduction,Signaling Pathway,Stem Cells,Surface Molecules,Transcription,Transcription Factors,Transcription Factors/Regulators.
    • Supports comparative expression analysis across conditions, genotypes, or treatments when paired with appropriate controls.
    • Useful for confirming target presence and subcellular distribution using orthogonal readouts (e.g., microscopy vs. immunoblotting).

    Common research applications

    • Western blot (WB): Compare relative target abundance and apparent size/isoforms across samples; interpret bands in light of expected MW and potential PTMs.
    • Immunohistochemistry (IHC): Assess tissue distribution and cell-type patterns; interpret staining with appropriate negative controls and antigen context.
    • Immunofluorescence / ICC: Visualize subcellular localization and co-localization patterns; consider fixation/permeabilization compatibility and controls.

    Notes for experimental interpretation

    • Consider isoforms, post-translational modifications, and processing that can shift apparent molecular weight or localization.
    • Use appropriate positive and negative controls (e.g., KO/KD, blocking peptide, or isotype controls) to support specificity interpretation.

As a monoclonal antibody, this reagent is expected to recognize a defined epitope, which can support consistency across lots when epitope accessibility is preserved.
