Product Overview
Clone IFA-19 is a rabbit monoclonal antibody that recognizes signal transducer and activator of transcription 3 (gene STAT3) phosphorylated at Tyr705 in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF and immunoprecipitation. The predicted molecular weight is 88.1 kDa, and the supplier reports an observed band at about 88 kDa.
STAT3 is a transcription factor activated mainly by IL-6-family cytokines through gp130 and JAK kinases, and also by growth factors and leptin. Phosphorylated STAT3 dimerizes, enters the nucleus and regulates genes for acute-phase responses, cell survival and proliferation. It shapes Th17 differentiation and is persistently active in many cancers, making it a major research target.

Samples with a confirmed band (WB): human HeLa cells, mouse NIH/3T3 cells, mouse RAW264.7 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:5000 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human lung cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): HeLa cells.
Immunogen
Synthetic peptide based on the sequence of human STAT3.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places signal transducer and activator of transcription 3 in the cytoplasm and nucleus. Tissue expression noted by UniProt (human): Heart, brain, placenta, lung, liver, skeletal muscle, kidney and pancreas. The samples tested by the supplier (listed above) are a practical starting point for positive controls. Because the antibody is phospho-specific, signal depends on the phosphorylation state of the sample; a phosphatase-treated lysate is a useful negative control.
