Product Overview

Clone EDD-19 is a rabbit monoclonal antibody that recognizes signal transducer and activator of transcription 5A (gene STAT5A) phosphorylated at Tyr694 in human and mouse samples and is validated for Western blot and IHC. The predicted molecular weight is 90.6 kDa, and the supplier reports an observed band at about 95 kDa.

STAT5A is a transcription factor activated by prolactin, growth hormone, IL-2, KIT ligand and other cytokines and growth factors. It binds GAS elements in target genes and is essential for prolactin-driven mammary gland development and milk protein expression, and it is also studied in hematopoiesis and lymphocyte signaling.

Validated Applications

Western blot 1:500–1:2000
Immunohistochemistry 1:50–1:200

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Samples with confirmed staining (IHC): human thyroid tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Immunogen

Synthetic phosphopeptide around Tyr694 of human stat5.

Reactivity Notes

The supplier lists reactivity with human and mouse. UniProt places signal transducer and activator of transcription 5A in the cytoplasm and nucleus. The samples tested by the supplier (listed above) are a practical starting point for positive controls. Because the antibody is phospho-specific, signal depends on the phosphorylation state of the sample; a phosphatase-treated lysate is a useful negative control.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunohistochemistry 1:50–1:200. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 90.6 kDa; observed band at about 95 kDa.
Q.What immunogen was used to raise this antibody?
A.Synthetic phosphopeptide around Tyr694 of human stat5.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
