Product Overview
This rabbit polyclonal antibody detects phosphatidylinositol 4-kinase beta (gene PI4KB) in human, mouse and rat samples and is validated for Western blot, IHC, flow cytometry and ELISA. The predicted molecular weight is 91.4 kDa, and the supplier reports an observed band at about 95 kDa.
PI4KB (PI4Kβ) is a type III phosphatidylinositol 4-kinase that generates PI4P mainly at the Golgi, supporting secretory traffic from the Golgi to the plasma membrane. It is also hijacked by several positive-strand RNA viruses, such as enteroviruses, to build their replication organelles, making it a target in both membrane biology and antiviral research.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human, Mouse, Rat) |
|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Human) |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human K562 cells, human HeLa cells, rat brain tissue, mouse brain tissue, mouse lung tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human skeletal muscle tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with a confirmed signal (flow cytometry): Jurkat cells, U251 cells.
Immunogen
Recombinant human PI4KB fragment (Asp375–Asp671), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 100% and 99.7%, respectively.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places phosphatidylinositol 4-kinase beta in the endomembrane system, mitochondrion outer membrane, rough endoplasmic reticulum membrane and Golgi apparatus. Tissue expression noted by UniProt (human): Widely expressed with highest levels in heart, skeletal muscle, pancreas, testis and ovary. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
